Go to Control Panel/Device Manager. Once in device manager, look for yellow question marks especially where the USB stuff is. It may be something simple as uninstalling and re-installing through device manager, or updating the driver.

One thing about this anytone at6666 got my DE-2000 hooked up want.to set the mod to check it Mic gain all the way open and on the meter says 50% hooked up my 959 opened mic gain all the way says 100% that's AM mod think it might need some adjustment but not sure which pot it is
